The coding strand mirrors the mRNA sequence but is not read; the template (non-coding) strand is complementary to the mRNA and is the one transcribed.

Of the two strands of a double-stranded DNA molecule, only one is used as the template for making a given mRNA, so the two strands have different, complementary roles:

  1. Coding strand (sense strand): this strand has a base sequence identical to that of the mRNA produced (except that DNA has thymine, T, where the mRNA has uracil, U). Because it carries the same sequence as the mRNA (and hence, indirectly, the sequence that will be translated into protein), it is called the coding strand -- but it is important to note that this strand itself is not the one directly copied by RNA polymerase during transcription. …
