Steroid hormones cross the membrane and act via intracellular receptors on DNA; proteinous hormones bind surface receptors and act via a second messenger.

Steroid and proteinous (protein/peptide) hormones act through two fundamentally different mechanisms, based on their solubility.

Step 1 (Steroid hormones). Being lipid-soluble, steroid hormones (and thyroid hormones) pass directly through the target cell's plasma membrane into the cytoplasm, where they bind a specific intracellular receptor protein, forming a hormone-receptor complex.

Step 2. This complex then enters the nucleus and binds a specific regulatory site on the DNA, activating transcription of specific genes into mRNA, which then directs new protein/enzyme synthesis in the cytoplasm -- action here is comparatively slow to start (since it depends on gene transcription and translation) but long-lasting.
