Capping and tailing are post-transcriptional modifications that add protective structures to the 5' and 3' ends of eukaryotic mRNA respectively.

In eukaryotes, the primary transcript (pre-mRNA/hnRNA) produced by RNA polymerase II undergoes several processing steps before it becomes a mature, functional mRNA that can be translated. Two of these steps are capping and tailing.

Capping occurs at the 5' end of the pre-mRNA, usually while transcription is still in progress. A specific enzyme adds a modified guanine nucleotide, 7-methylguanosine (m7G), to the first transcribed nucleotide. Unlike the normal 5'-3' phosphodiester bonds found elsewhere in the RNA chain, this cap is attached through an unusual 5'-5' triphosphate linkage. The cap serves several important functions: it protects the mRNA from degradation by 5' exonucleases, is required for efficient export of the mRNA from the nucleus to the cytoplasm, and is recognised by initiation factors that help the ribosome bind correctly to begin translation.
