Missense mutation changes one amino acid in the protein; nonsense mutation prematurely terminates translation.

Both missense and nonsense mutations are types of point mutations (base-pair substitutions) that alter a single codon in the DNA/mRNA sequence, but their effects on the resulting protein are very different.

A missense mutation occurs when the base change alters a codon so that it now specifies a different amino acid than the one originally coded for. The resulting protein is still full length, but carries one incorrect amino acid at that position. Depending on where the substitution occurs and how chemically similar the new amino acid is to the original, the protein's function may be unaffected, reduced, or completely lost. A well-known example is the mutation in the beta-globin gene that replaces glutamic acid with valine, producing abnormal haemoglobin and causing sickle-cell anaemia.
