The first two codon-anticodon base pairs are always strict; the third position tolerates non-standard 'wobble' pairs, letting one tRNA read more than one codon.

During translation, the anticodon of tRNA pairs antiparallel with the mRNA codon. At the first two codon positions, pairing strictly follows Watson-Crick rules (A-U and G-C). For example, in perfect pairing, codon 5'-GCA-3' pairs with anticodon 3'-CGU-5' at all three positions. However, Francis Crick's wobble hypothesis explains that at the third codon position (which pairs with the first, 5' base of the anticodon), the geometric constraints are relaxed, permitting some non-standard base pairs.
