During daylight hours, microfilariae remain concentrated in the deep blood vessels of the body's internal organs, particularly the lungs, and are largely undetectable in blood taken from a peripheral site such as an arm vein. At night, however, large numbers of microfilariae migrate out of these deep vessels into the peripheral circulation, becoming readily detectable in a blood smear taken at that time. This rhythm closely tracks the biting behaviour of the Culex mosquito, the vector responsible for transmitting the parasite, which feeds predominantly during the night — so that microfilariae are present in peripheral blood exactly when a mosquito is most likely to take a blood meal and pick them up.
