The HSK/C4 pathway pre-concentrates CO2 in mesophyll cells before passing it to bundle-sheath cells for the Calvin cycle, boosting photosynthetic efficiency in hot, dry climates.

The Hatch-Slack-Kortschak (HSK) pathway, also called the C4 pathway, operates in plants like maize, sugarcane and sorghum that show Kranz anatomy. Step 1: in the mesophyll cells, atmospheric CO2 (as HCO3-) combines with the 3-carbon acceptor phosphoenolpyruvate (PEP), catalysed by PEP carboxylase (an enzyme with very high affinity for CO2 and no oxygenase activity), forming the 4-carbon compound oxaloacetic acid (OAA). Step 2: OAA is quickly reduced to malic acid (or transaminated to aspartic acid) in the mesophyll chloroplast. Step 3: this 4-carbon acid is transported through plasmodesmata into the neighbouring bundle-sheath cells. Step 4: there it is decarboxylated, releasing CO2 (raising local CO2 concentration sharply) and forming pyruvic acid. Step 5: the released CO2 now enters the ordinary Calvin cycle (C3 pathway) inside the bundle-sheath chloroplasts, being fixed by RuBisCO to eventually form sugars. Step 6: the pyruvate p …
